内耳相关性BDNF和NT-3的研究进展

神经营养因子(neurotrophin)是一组与神经结构和功能密切相关的家族蛋白,它包括神经生长因子(NGF)、脑源性神经营养因子(BDNF)、胶质细胞源性神经营养因子(GDNF)、神经营养素-3(NT-3)、神经营养素-4/5(NT-4/5)以及最近发现的NT-6.神经营养因子是外周和中枢神经系统中最重要的内源性蛋白,调控着神经元的发育、生长、分化和生存,对维持神经系统的正常功能承担着重要作用.随着近些年对神经营养因子的进一步研究认识和基因转染的实验进展,以BDNF和NT-3为代表的神经营养因子正在被越来越多的研究人员用来实验治疗动物的各种先天性神经系统疾病.过去一般认为在发育的早期阶段,靶源性的神经营养因子是神经元生存所必需的,这种依赖性是随着成熟而减少,至成年时期逐渐消失.但是近些年的研究证据表明:某些神经元在成年期的生存仍然需要营养信号,颅部的感觉神经特别是耳蜗-前庭系统也不例外[1]. ……
